1) Evaluation of FOXP3 gene polymorphisms in patients with allergic disease compared with controls

  • DNA extraction from 200 allergic patient samples and 200 normal samples
  • Designing Primers for amplification of two regions within FoxP3 gene
  • PCR amplification of the two regions of foxp3 gene
  • Analysis of the PCR product via DNA electrophoresis
  • Analyzing single nucleotide polymorphism associated with the FoxP3 gene via Restriction Fragment Length Polymorphism (RFLP) test
  • Confirming the RFLP results via sequencing analysis
  • Statistical data analysis via SPSS software

2) Identification, isolation and evaluation of prostate-specific promoters for gene therapy

  • Exploring the NCBI, Unigene and, other expression databases to identify the favorable prostate specific promoters
  • Designing primers for specific prostate promoters
  • Isolating the target regions via PCR amplification
  • Analysis of the PCR product via DNA electrophoresis Cloning the aforementioned promoters into suitable expression vectors
  • Transfecting vectors into prostate and several other cell lines using PEI
  • Examining the transfection efficiency via fluorescence microscopy
  • Determining the transfection efficiency using flow cytometry
  • Substituting the eGFP gene by thymidine kinase as a requirement of downstream application in gene therapy
  • Examining cytotoxicity via tunnel assay
  • Determining the level of thymidine kinase protein expression via real-time PCR
